The Matrix Reloaded: New Insights from Type IV Collagen Derived Endogenous Angiogenesis Inhibitors and their Mechanism of Action
نویسنده
چکیده
Angiogenesis, the process of neovascularization from parent blood vessels, is a prerequisite for many physiological and pathological conditions that is regulated by a balance between the levels of endogenous angiogenic stimulators and matrix reloaded angiogenic regulators. Several non-collagenous carboxy terminal end domains in chains of type IV collagen matrix reloaded molecules selectively interact with proliferating endothelial cells by binding to distinct integrins and regulate intracellular signaling and inhibit angiogenesis. This review will focus on the current understanding of extra cellular matrix type IV collagen reloaded endogenous angiogenesis inhibitors and their mechanism of action.

Signaling Mechanisms of Endogenous Angiogenesis Inhibitors Derived from Type IV Collagen
Vascular basement membrane (VBM) derived molecules are regulators of certain biological activities such as cell growth, differentiation and angiogenesis. Angiogenesis is regulated by a systematic controlled balance between VBM derived antiangiogenic factors and proangiogenic growth factors. In the normal physiological state, equilibrium is maintained between the antiangiogenic and proangiogenic...
متن کاملChanging Roles of Matrix Metalloproteases and Their Inhibitors, TIMPs, During Tumor Progression and Angiogenesis
Inhibition of matrix-metalloproteinases (MMPs) by tissue inhibitors of metalloproteinases (TIMPs) has been shown in vivo to decrease metastasis and tumor-associated angiogenesis. Our laboratory is interested in understanding the role of these proteins at the pericellular microenvironment of tumor and endothelial cells. Secretion of MMPs by tumor cells enables the migration, invasion and metasta...
متن کاملThe role of matrix metalloproteases and their inhibitors in tumour invasion, metastasis and angiogenesis.
One critical event of tumour invasion that signals the initiation of the metastatic cascade is thought to be interaction of the tumour cell with the basement membrane. Basement membranes may also pose as barriers to tumour cell invasion at multiple points later in the metastatic cascade, including during the processes of vascular infiltration and extravasation. Thus, an important proteolytic ev...

Several types of collagen contain cryptic antiangiogenic noncollagenous domains that are released upon proteolysis of extracellular matrix (ECM). Among those is Arresten, a collagen-derived antiangiogenic factor (CDAF) that is processed from a1 collagen IV. However, the conditions under which Arresten is released from collagen IV in vivo or whether the protein functions in tumor suppressor path...
